Orocyan®: a cranberry extract rich in A-type proanthocyanidins

Orocyan® is a cranberry extract (Vaccinium macrocarpum) rich in A-type proanthocyanidins, a berry flavonoid.

Orocyan® has anti-infective properties and may have a beneficial role in weight management and in metabolic syndrome.

Orocyan® is available in Benelux countries and in Spain.

Description

Orocyan® is a hydroalcoholic extract of the cranberry (Vaccinium  macrocarpum) with concentrations of 20% proanthocyanidins (HPLC assay) and 10% proanthocyanidins (type A2 – DMAC assay).

Orocyan® has antioxidant and anti-inflammatory activities and is a good anti-infective agent for urinary infections.

Orocyan : anti-infective properties for urinary health

We use to use cranberry to fight urinary tract infection.

According to Nicolasi (2014), acute uncomplicated cystitis (AUC) also called urinary tract infection  occurs when urinary pathogens from the bowel or vagina ( for women ) colonize the periurethral mucosa and reach the bladder.About 90% of AUC are caused by uropathogenic Escherichia coli (UPEC).

Di Martino et al. conducted a randomized double-blind placebo-controlled study with 32 subjects. This study has shown a significant urinary anti-adhesion effect against bacteria.

This activity has been highlighted thanks to urine samples collected from volunteers that consumed cranberry powder compared to placebo (p < 0.001). This inhibition was clearly dose-dependent, prolonged (until 24 h with 72 mg of PAC).

A supplementation  of cranberry extract could protect urinary tract against cells bacterian adhesion. 

Into the in vitro study of  Nicolisi et al (2014), the aim was to evaluate the inhibiting activity of type A2 proanthocyanidins on strains of E. coli , isolated from urine and responsible for cystitis.

The adhesion assay was carried out with six groups of contaminated cells and one control group. 5 different strains of E. coli were isolated from the urine of women with a history of recurrent cystitis. The cells were grown on a sterile cover glass and then with the addition of type A proanthocyanidins, at different concentrations from 25 µg/mL to 100 µg/mL. The cells were then colored with and observed at the microscope. The adhesion index obtained is the average number of adhering bacteria per cell, from an examination of 100 cells.

The figure below illustrates the adhesion index depending on the different strains of E. Coli and concentrations of cranberry extracts.

Urinary health

The adhesion index decreased. For the strain n° 1, the adhesion index was 74.5% in the treated group whereas it was 12.3% in the control group with a concentration of 100 µg/ mL. The cranberry extract allows a reduction of the adhesion of E. coli to the cells and this adhesion is done in a dose-dependent manner.

The cranberry extract can then reduce the ability of E. coli to adhere to the uroepithelial cells and infect the bladder.

Weight management and metabolic syndrome

Cranberry extract can be beneficial for health beyond urinary health.

Some beneficial results have been shown on obesity in an animal study (C57BL/6J mice).(Anhê, 2015).  The aim of the study is to investigate the metabolic impact of a cranberry extract (CE) on high fat/high sucrose (HFHS)-fed mice, which induces a weight gain and obesity.

There were 3 groups. On group was fed daily with a high-fat high-sugar diet (HFHS). The second group was fed with the HFHS diet and with a cranberry extract (200mg/kg). The third group was the control group fed with a normal diet and water for 8 weeks. The cranberry extract (CE) supplementation has reduced the weight gain and the energy efficiency (g/kcal* 1000).

CE supplementation has also decreased triglyceride accumulation and some markers of oxidative stress, inflammation and has improved insulin sensitivity. It may decrease the proportion of Akkermansia spp., which has a key role in lipidic and glucidic metabolism. A low rate is often observed in obese patients or patients suffering from metabolic syndrome.

Thus, Orocyan could play a role in weight management.

Regulatory Information

Orocyan® is a fruit (cranberry) extract authorized for use in food supplements in France.

In France, Vaccinium macrocarpum is listed in Annex I of the French “Plants” decree of 24 June 2014 establishing the list of plants authorized for use in food supplements and the conditions of their employment (authorized plant parts: fruit, leaves).

In Belgium, V. macrocarpum is listed in the Belgian Royal Decree of 29 August 1997 on plants on the  manufacture and marketing of foodstuffs constituted or consisting of plants or plant preparations (authorized plant part: berry).

Cranberries, being consumed in Europe since before May 1997, are not subject to Regulations (EU) nos. 2015/2283 and 258/97 on novel foods.
